Abstract:

摘要 目的:探讨磷酸二酯酶4D(PDE4D)在口腔鳞状细胞癌(OSCC)发生发展过程中的作用。方法:收集53例OSCC患者标本和5种OSCC细胞株,55例正常健康标本及2种正常口腔上皮细胞作为对照,利用qRT-PCR检测其中PDE4D mRNA表达水平;分别利用CCK-8,细胞划痕实验,流式细胞术和Western blot等技术检测PDE4D抑制剂Rolipram对SSC25细胞增殖、迁移、凋亡的影响。结果:OSCC患者组织中PDE4D mRNA相对水平显著高于正常对照组;OSCC细胞株中PDE4D mRNA相对水平显著高于正常细胞组,差异有显著统计学意义(P<0.01);且分化程度越高,PDE4D mRNA相对水平越高(P<0.05);Rolipram显著抑制SSC25细胞的增殖能力和迁移能力,促进细胞凋亡(P<0.05)。结论:PDE4D在OSCC发生发展过程中起关键调控作用
