14,15-环氧二十碳三烯酸促进颅骨缺损骨再生

Abstract:
摘要 目的: 建立C57BL/6小鼠颅骨缺损模型,研究14,15-环氧二十碳三烯酸(14,15-epoxyeicosatrienoic acid,14,15-EET)能否促进骨再生,为骨组织再生研究提供新思路。方法: C57BL/6小鼠颅中缝两侧建立直径5 mm的全层颅骨圆形缺损,小鼠右侧(实验侧)缺损处植入14,15-EET预处理的骨髓间充质干细胞(BMSCs),左侧(对照侧)植入未用14,15-EET处理的BMSCs。术后第3天于实验侧缺损处局部皮下注射含14,15-EET(10 μmol/L)磷酸缓冲盐溶液(PBS),对照侧缺损处皮下注射等量PBS,隔日注射,持续4周。术后8周将小鼠处死,CBCT放射诊断观测缺损面积大小,HE染色和Masson三色染色分析新骨形成相关组织结构。结果: 术后8周小鼠实验侧缺损面积比对照侧明显减小,差异有统计学意义(P<0.05);实验侧缺损区新生血管数、新生骨量均较对照侧明显增多,差异有统计学意义(P<0.05);此外,实验侧颅骨缺损区可明显观察到骨质及胶原纤维,而对照侧不明显。结论: 通过局部提高14,15-EET水平可促进新生骨形成,推测可能与14,15-EET改善局部微循环相关
