Abstract:

摘要 目的:观察槲皮素对ACC-M细胞裸鼠移植瘤的抑制作用,探讨其作用机制。方法:常规将ACC-M 细胞接种于裸鼠右侧背部皮下,待肿瘤长至直径约6mm时随机分为5组,即空白对照组、槲皮素低、中、高剂量组、紫杉醇组,每组6只。定期测量肿瘤体积,绘制肿瘤生长曲线。给药3周后处死裸鼠,剥取肿瘤,称瘤重,计算抑瘤率。苏木精-伊红(hematoxylin-eosin,HE)染色,免疫组织化学法检测血管内皮生长因子(vascular endothelial growth factor,VEGF)和增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)的表达情况。结果:给药3周后,各组的抑瘤率分别为11.45%、37.67%、42.49%、43.46%。VEGF表达的平均吸光度值分别为(0.420±0.039)、(0.367±0.040)、(0.315±0.033)、(0.270±0.016)、(0.298±0.044),各组比较差异有统计学意义(P<0.05);PCNA阳性表达率分别为(68.45±3.05)%、(58.84±3.33)%、(49.83±1.81)%、(44.90±1.85)%、(41.23±1.04)%,各组比较差异有统计学意义(P<0.01)。结论:槲皮素可抑制 ACC-M 细胞在裸鼠体内增殖,其机制可能通过抑制 VEGF和PCNA的表达发挥作用
