SRSF3调控口腔癌细胞中SPAG5外显子3的可变剪切的研究

Abstract:

摘要 目的:研究调控前体mRNA(pre-mRNA)可变剪切的原癌基因SRSF3对细胞有丝分裂的重要功能蛋白SPAG5的外显子3的可变剪切的调控作用。方法:在人口腔鳞状上皮细胞癌细胞系CAL 27中应用RNAi技术降低SRSF3的表达,通过逆转录PCR(RT-PCR)检测SPAG5外显子3的可变剪切。结果:在CAL 27细胞中抑制SRSF3表达时,抑制了全长的SPAG5的表达,增加了较短的SPAG5的剪切异构体的表达。结论:SRSF3可以调控SPAG5外显子3的可变剪切,并可能影响了SPAG5蛋白的表达水平和有丝分裂
