Abstract:

摘要 目的:评价甲状旁腺激素融合蛋白PTH-FC(parathyroid hormone -Fc)对大鼠骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)骨向分化的作用,筛选出PTH-FC诱导BMSCs最佳的作用时间。方法:采用全骨髓贴壁法分离培养大鼠BMSCs并进行鉴定,以1.0×10-9 mol/L PTH-FC成骨诱导液诱导BMSCs作用6 h(间歇性给药组)和48 h(持续性给药组)通过噻唑蓝(methylthiazolyldiphenyl-tetrazolium bromide, MTT)、碱性磷酸酶(alkaline phosphatase,ALP)、茜素红染色观察PTH-FC对BMSCs增殖和骨向分化的作用。结果:含1.0×10-9 mol/L PTH-FC的成骨培养液作用6 h间歇性给药组与其他组比较,ALP和茜素红染色结果显著高于其他组;MTT结果显示各实验组间比较差异无统计学意义。结论:1.0×10-9 mol/L PTH-FC作用BMSCs 6h的间歇性给药组具有最佳的骨向诱导能力,这提示PTH-FC作为骨诱导生长因子联合BMSCs对骨组织缺损的修复是可行的
