Abstract:

摘要 目的:研究牙龈干细胞(Mesenchymal stem cells derived from gingiva, GMSCs)对小鼠实验性自身免疫性脑脊髓炎 (experimental autoimmune encephalomyelitis, EAE) 的治疗作用,为干细胞治疗自身免疫性脑脊髓炎提供实验依据。方法:通过GMSCs与T细胞体外共培养,研究GMSCs对T细胞凋亡、增殖以及向Treg细胞和Th17细胞分化的影响。建立小鼠EAE模型,研究GMSCs对EAE的治疗作用。结果:GMSCs在体外共培养中诱导T细胞凋亡, 同时抑制T细胞增殖。GMSCs可以促进Treg细胞分化,而抑制Th17细胞分化。GMSCs注射对小鼠EAE具有治疗作用。机制上GMSCs注射促进EAE小鼠中Treg细胞分化,抑制Th17细胞分化,抑制炎症反应。结论:GMSCs通过促进Treg细胞分化,抑制Th17细胞分化对小鼠EAE具有治疗作用
