不同锌含量Zn/PLGA/HA复合支架对人牙髓干细胞粘附能力影响的体外研究

Abstract:

摘要 目的:制备含有不同锌含量的Zn/PLGA/HA复合支架并检测其机械性能;探讨不同锌含量的Zn/PLGA/HA复合支架在体外对人牙髓干细胞粘附能力的影响。方法:通过溶液浇铸/粒子沥滤法制备含锌质量分数0.5%、1%及2%的Zn/PLGA/HA复合支架,每组HA含量均为20%质量分数,对照组为不含锌的20%HA/PLGA支架。使用扫描电镜观察支架的表面形态,电子万能试验机检测支架的拉伸强度。将人牙髓干细胞接种在不同锌含量的Zn/PLGA/HA复合支架上培养,采用DAPI染色细胞计数法检测细胞粘附能力。结果:含锌质量分数1%和2%的Zn/PLGA/HA复合支架组拉伸强度均高于对照组(P<0.05)。扫描电镜观察到各组支架表面孔隙率均较高(87.6%),孔径158~372 μm,并具有良好的孔间连通性。0.5%Zn组支架与对照组相比具有较好的促牙髓干细胞粘附能力(P<0.05)。结论:本实验中,含1%,2% Zn(wt%)的Zn/PLGA/HA支架具有较好的机械性能,含0.5%Zn(wt%)的Zn/PLGA/HA支架在体外对人牙髓干细胞粘附具有更好的促进作用,为含锌支架作为支架材料用于组织工程提供了理论基础
