华卟啉钠介导的光动力疗法对人舌鳞癌细胞株HSC-4的增殖和迁移能力的影响

Abstract:

摘要 目的:探讨华卟啉钠(sinoporphyrin sodium,DVDMS)介导的光动力疗法(photodynamic therapy,PDT)对体外培养的口腔舌鳞状细胞癌(oral tongue squamous cell carcinoma,OTSCC,简称鳞癌)细胞株HSC-4的增殖及迁移能力的影响。方法:检测DVDMS细胞毒性,细胞摄取药物情况,将细胞分为对照组以及不同浓度的DVDMS-PDT处理组(DVDMS孵育浓度分别为0.1、0.5、1.0、2.0、3.0、4.0、5.0、6.0、8.0 mg/L),激光能量密度为0.75、1.50、3.00、6.00 J/cm2,细胞增殖检测试剂盒(cell counting kit-8,CCK-8)法检测DVDMS-PDT对细胞增殖能力的影响,划痕实验和Transwell小室实验观察细胞迁移能力的变化。结果:DVDMS毒性实验显示单纯药物对细胞毒性较小,联合激光照射后,可显著抑制HSC-4细胞增殖(P均<0.05),与对照组比较,划痕愈合率降低,Transwell小室穿膜细胞数减少(P均<0.05)。DVDMS药物浓度2.0 mg/L,体外孵育6 h,光能量密度在3.00 J/cm2以上可有效杀伤HSC-4。结论:DVDMS-PDT可有效杀伤舌鳞癌细胞,抑制HSC-4的迁移。光敏剂浓度、孵育时间、光能量密度是影响疗效的重要因素
