Abstract:

摘要 目的:研究浓缩生长因子对成骨细胞骨唾液酸蛋白(bone sialoprotein,BSP)、runt转录相关因子2(runt related transcription factor 2,Runx2)、碱性磷酸酶(alkaline phosphatase,ALP)基因表达的影响,探讨浓缩生长因子促进大鼠种植体骨结合的作用机制。方法:采用噻唑蓝比色法(methylthiazolyldiphenyl-tetrazolium bromide,MTT)和实时定量反转录聚合酶链反应(real-time reverse transcriptase-polymerase chain reaction,RT-qPCR)法检测浓缩生长因子对成骨细胞增殖和BSP、Runx2、ALP基因表达的影响;建立大鼠股骨种植体骨结合模型,应用Micro-CT扫描测定标本骨矿密度、骨体积百分比等数据。结果: 浓缩生长因子可以促进成骨细胞的增殖和成骨分化相关基因BSP、Runx2、ALP的表达。种植体植入8周后,与对照组比较,浓缩生长因子组骨矿密度、骨体积百分比、骨小梁数量和成骨界面面积均显著升高。结论: 浓缩生长因子可能通过增加成骨相关基因表达而促进大鼠种植体骨结合,浓缩生长因子是一种安全、无排斥反应的理想生物膜材料
