唑来膦酸对牙龈成纤维细胞生物学特性的影响研究

Abstract:
摘要 目的:探讨唑来膦酸对牙龈成纤维细胞增殖、凋亡、分化的影响及机制。方法:从牙龈组织分离原代人牙龈成纤维细胞(HGFs),0.5、1、5、10 μmol/L的唑来膦酸处理HGFs 24、48、72 h后,CCK8实验检测细胞的增殖;流式细胞仪检测10 μmol/L的唑来膦酸处理HGFs 72 h后的细胞凋亡情况;加入10 μmol/L的唑来膦酸的成骨诱导分化液培养HGFs 7 d后,碱性磷酸酶(ALP)试剂盒检测ALP活性,Western blot检测Runx2和OPN的蛋白表达;10 μmol/L的唑来膦酸处理HGFs 72 h后,Western blot检测NF-κB信号通路p65、IκBα蛋白表达。结果:唑来膦酸可呈剂量依赖抑制HGFs细胞增殖(P<0.01);唑来膦酸组细胞的凋亡率显著高于对照组,ALP活性及Runx2、OPN蛋白表达显著高于对照组,p65、IκBα的蛋白表达显著低于对照组(P<0.01)。结论:唑来膦酸可抑制HGFs增殖,促进细胞的凋亡和分化,其机制与下调NF-κB信号通路有关
