不同频率电刺激对压力性尿失禁模型小鼠盆底组织胶原代谢的影响 Effects of Electrical Stimulation at Different Frequencies on Metabolism of Collagen in Pelvic Supportive Tissue

Abstract:

目的:探讨电刺激对阴道扩张法(VD)诱导的压力性尿失禁(SUI)模型小鼠盆底组织胶原代谢的影响。方法:对SUI模型小鼠施加7d20Hz或50Hz频率电刺激(ES)后,测定尿动力学指标评价造模效果、采用免疫荧光双标法和Western-blot法检测电刺激前后小鼠盆底组织Ⅰ型胶原(ColⅠ)和Ⅲ型胶原(ColⅢ)含量变化。结果:与对照组相比,VD组小鼠的最大膀胱容量(MBC)和漏尿点压(LPPs)值明显增高(P<0.001),ColⅠ和ColⅢ相对荧光值和蛋白表达明显降低(P<0.001),且Ⅰ/Ⅲ胶原比降低,胶原形态疏松紊乱;与VD组比较,VD+ES处理之后小鼠MBC和LPPs值明显增高(P<0.01,P<0.001),ColⅠ和ColⅢ相对荧光值明显增加(P<0.05,P<0.01)、Ⅰ/Ⅲ胶原比升高(P<0.01,P<0.001)其中VD+ES50组相对荧光值和蛋白表达增加量(P<0.01)较VD+ES20组(P<0.05)多。结论:电刺激治疗能有效改善SUI模型鼠膀胱储尿功能及尿道控尿功能,其中50 Hz的高频电刺激效果更为显著,其机制可能与促进小鼠盆底组织Ⅰ、Ⅲ型胶原的代谢有关
