SMYD2在子宫内膜异位症中的作用 Role of SMYD2 in Endometriosis

Abstract:

目的:探讨组蛋白甲基转移酶(SMYD2)在子宫内膜异位症(EMT)患者子宫内膜组织及基质细胞中的表达及其对p53、白血病抑制因子(LIF)的调节作用。方法:体外诱导对照组和EMT组子宫内膜基质细胞蜕膜化,蜕膜化同时应用SMYD2特异性抑制剂AZ505,即分为空白组、蜕膜化组、蜕膜化+抑制剂组,采用Real-time PCR法和细胞免疫化学法检测SMYD2,转化相关蛋白53(TRp53)mRNA和蛋白在子宫内膜蜕膜化细胞的表达及SMYD2特异性抑制剂AZ505对其表达的影响,并通过检测对照组和EMT组各时期子宫内膜组织中SMYD2、TRp53mRNA和蛋白的表达规律,从而探究SMYD2在EMT中的作用。结果:SMYD2、TRp53mRNA在对照组子宫内膜各时期均有表达,其中在分泌中期表达下降,而且SMYD2mRNA在EMT组子宫内膜分泌中期表达较对照组下降,免疫荧光法可见SMYD2、p53和p-p53蛋白在分泌中期信号减弱,在EMT组MSP期SMYD2、p53和p-p53蛋白较对照组MSP期增强。SMYD2、TRp53mRNA和蛋白的表达在对照组和EMT组基质细胞蜕膜化过程中均较各自空白组降低,应用SMYD2特异性抑制剂AZ505后,对照组细胞中SMYD2mRNA与其蜕膜化组比较差异无统计学意义,TRp53mRNA较其蜕膜化组降低,EMT组SMYD2、TRp53mRNA与其蜕膜化组差异无统计学意义;对照组和EMT组SMYD2和p53、p-p53蛋白信号均较蜕膜化细胞减弱。结论:SMYD2对子宫内膜中p53和LIF有调节作用
