Abstract:

目的:探讨温度敏感性水凝胶Dex-PCL-HEMA/PNIPAAm对大鼠心肌缺血再灌注损伤的作用。方法:结扎SD大鼠冠状动脉左前降支30min后放松结扎线,制作心肌缺血再灌注(IR)模型,以仅穿线但不结扎冠状动脉者为模型对照组(Sham组)。将IR模型制作成功的大鼠随机分为2组:磷酸盐缓冲溶液组(PBS组):缺血周边区注射PBS;水凝胶组(Gel组):缺血周边区注射Dex-PCL-HEMA/PNIPAAm水凝胶。28d后行超声心动图、组织病理学及免疫组织化学检测。结果:与PBS组相比,Gel组左室舒张末内径(LVEDd)、左室收缩末内径(LVESd)减小,左室射血分数(LVEF)升高,心肌细胞凋亡指数降低,梗死减小,梗死区室壁厚度增加(P<0.05)。结论:心肌IR后缺血周边区注射Dex-PCL-HEMA/PNIPAAm水凝胶可抑制细胞凋亡、减少梗死面积、保护IR后大鼠心功能
