人膀胱逼尿肌肌球蛋白的表达及体外功能研究 Expression and Functional Activities of Smooth Muscle Myosin and Non-Muscle Myosin Isoforms in Human Bladder Detrusor Urinae

Abstract:

目的:探讨平滑肌(SM)肌球蛋白(SMM)及非肌肌球蛋白(NMM)在人膀胱逼尿肌中的表达及功能。方法:人膀胱逼尿肌标本(n=13)来自于武汉大学中南医院心脏死亡患者捐献的、病理证实为正常膀胱逼尿肌组织的标本;Organ Bath检测膀胱逼尿肌收缩和舒张特性;竞争性RT-PCR检测SMM同工异构体(SM-A/-B,肌球蛋白轻链MLC17a/b和SM1/2)的表达;Western Blot和免疫荧光染色对膀胱SMM重链(MHC)和NMM重链的三种同工异构体(NA,NB和NC)进行检测并定位。结果:Carbachol可以显著诱导膀胱逼尿肌收缩,表现为典型的时相收缩特性;膀胱逼尿肌表达约20%SM-B,70%MLC17a和45%SM2;此外,NMM重链的三种同工异构体(NA,NB和NC)广泛存在于膀胱SM和内皮细胞中,而MHC则仅表达在膀胱及血管SM中。同时,肌球蛋白选择性抑制剂Blebbistatin可以显著舒张苯肾上腺素预收缩的膀胱逼尿肌。结论:SMM及NMM同工异构体在膀胱逼尿肌收缩舒张中可能起到重要的作用
