Abstract:

目的:探讨USP22基因在氟尿嘧啶对肝癌细胞毒性作用中的影响。方法:用Western blotting检测肝癌细胞HepG2和BEL7402中USP22基因的表达状态,构建并对肝癌细胞转染USP22基因的siRNA(siRNAUSP22),然后以氟尿嘧啶处理24h后,MTT检测法观察在USP22基因沉默前后,氟尿嘧啶对肝癌细胞活性的影响。结果:USP22基因在肝癌细胞中显著高表达。siRNA-USP22转染后显著抑制USP22基因的表达,同时可明显增强氟尿嘧啶对HepG2和BEL7402细胞的毒性作用,降低其细胞活性[(53.2±6.8)%,P<0.05;(49.8±7.3)%,P<0.05]结论:下调USP22基因可提高肝癌细胞对氟尿嘧啶的敏感性,提示USP22高表达可能是造成肝癌化疗抵抗的原因之一
