抗氧化剂N-乙酰半胱氨酸对早期糖尿病大鼠心肌Caveolin-3/eNOS/NO信号通路的影响 Effects of Antioxidant N-Acetylcysteine on Caveolin-3/eNOS/NO Signaling Pathway in Early Diabetic Rats

Abstract:

目的:探讨抗氧化剂N-乙酰半胱氨酸(NAC)对早期糖尿病大鼠心肌Caveolin-3/eNOS/NO信号通路及心肌功能的影响。方法:清洁级健康成年雄性SD大鼠24只,随机分为对照组(NC组)、糖尿病组(DM组)和NAC治疗组(DM+NAC组),每组8只。大鼠经尾静脉一次注射链脲佐菌素60mg/kg造模。糖尿病模型制备成功1周后,DM+NAC组给予NAC 1.5g/(kg·d)灌胃治疗,NC组和DM组分别给予等量生理盐水,持续4周后超声检测心脏功能。试剂盒检测血清脂质过氧化特异性指标15-F2t-异前列烷(15-F2t-isoprostane)、超氧化物歧化酶(SOD)及总抗氧化物浓度水平,并检测心肌组织15-F2t-isoprostane、超氧阴离子(O_2~-·)、硝基酪氨酸(Nitrotyrosine)及一氧化氮(NO)水平;Western Blot法检测心肌小窝蛋白-3(Caveolin-3)、内皮型一氧化氮合酶(eNOS)及peNOS蛋白的表达。结果:与NC组比较,DM大鼠血清15-F2t-isoprostane水平与总抗氧化物浓度显著升高,而SOD活性显著降低(P<0.05);心肌组织15-F2t-isoprostane、O_2~-·与Nitrotyrosine水平明显升高,而心肌Caveolin-3、p-eNOS表达水平及NO水平明显降低(P<0.05),心肌舒张功能降低而心肌收缩功能未受影响。与DM组比较,NAC组血清15-F2t-isoprostane与总抗氧化物浓度水平明显降低,SOD及心肌NO水平明显升高(P<0.05);心肌组织15-F2t-isoprostane、O_2~-·及Nitrotyrosine水平明显下降,Caveolin-3与p-eNOS表达水平显著增加(P<0.05),心肌舒张功能改善。结论:早期糖尿病大鼠心肌氧化应激水平增加可能是Caveolin-3与p-eNOS表达水平下降,心肌舒张功能受损的原因之一。减轻氧化应激对Caveolin-3/eNOS/NO信号通路的影响,可能是抗氧化剂NAC改善糖尿病心肌舒张功能的另一重要机制
