Abstract:

目的:探讨骨髓间充质干细胞(MSC)对单核细胞分化为树突状细胞(DC)的影响。方法:将分离培养的MSC与单核细胞共培养,加入DC分化诱导因子GM-CSF和IL-4(MSC+-DC),并以培养分化体系中无MSC存在的作为阳性对照(MSC--DC)。6d后收集悬浮细胞并用TNF-α诱导成熟48h。分别用流式细胞术、混合淋巴细胞反应及扫描电子显微镜检测细胞的表型、免疫功能及细胞形态。结果:同阳性对照DC相比,与MSC共培养得到的细胞上DC相关分子的表达明显较低,包括CD80,CD86,CD1a,CD83;而CD14分子则持续性高表达;与MSC共培养的细胞刺激异基因T淋巴细胞增殖的能力亦显著降低。形态学上,细胞胞体小,胞质突起不明显甚至缺如。结论:MSC抑制单核细胞向DC分化和成熟
