MiR205在脓毒症小鼠心脑肾中对HMGB1表达的影响 Influence of miR205 on HMGB1 in the Brains,Hearts and Kidneys of Mice with Sepsis

Abstract:

目的:检测在胆碱能抗炎通路(CAP)中miR-205表达变化和对脓毒症小鼠脑、心、肾组织中高迁移率族蛋白B1(HMGB1)的调节作用。方法:15只BALB/C小鼠被随机分成3组:脓毒症组和CAP组各6只,对照(control)组3只,分别腹腔注射脂多糖(LPS)15mg/kg+生理盐水,LPS 15mg/kg+GTS-21 4mg/kg,等量生理盐水。24h后处死小鼠取材心、肾、脑组织。定量逆转录聚合酶链反应(qRT-PCR)和酶联免疫吸附试验(ELISA)检测脓毒症小鼠和胆碱能抗炎通路小鼠的脑、心、肾组织中miR-205与HMGB1的表达。结果:脓毒症小鼠脑、心、肾组织的miR-205表达与对照组相比无明显变化(P_2、P_4、P_6>0.05),CAP组均较其他两组明显升高(P_1、P_3、P_5<0.001)。脑、肾组织中HMGB1的表达CAP组较脓毒症组有明显降低(P_1、P_3<0.001)。心肌组织中HMGB1的表达在脓毒症组和CAP组差异无显著性(P5>0.05)。结论:激活胆碱能抗炎通路时,脑、心、肾组织中表达升高的miR-205降低脑、肾组织中HMGB1的表达
