家鸡钠尿肽受体B基因的克隆和组织表达分析

中文摘要:钠尿肽受体B(NPR-B)作为C-型钠尿肽(CNP)的选择性受体,可介导CNP的多种生理效应,包括调控垂体激素释放、维持心血管系统稳定、调节神经系统发育、调控细胞增殖及促进骨骼生长等。本研究以家鸡 Gallus gallus domesticus为模型,首次报道了 NPR-B基因的全长cDNA,并检测其在成体家鸡中的组织表达图谱。结果显示:家鸡 NPR-B基因cDNA全长为3 189 bp,可编码1 062个氨基酸。家鸡NPR-B与人 Homo sapiens、大鼠 Rattus norvegicus、非洲爪蟾 Xenopus laevis和斑马鱼 Danio rerio分别具有79%、78%、73%、67%的氨基酸序列一致性;同时采用实时荧光定量PCR技术探究家鸡 NPR-B基因的组织表达分布,发现其在心脏、肌肉、中脑和垂体(头部)中表达水平较高。本研究结果为阐释CNP及其选择性受体NPR-B在家鸡中的生理功能奠定基础。
英文摘要:As a selective receptor of C-type natriuretic peptide (CNP), the natriuretic peptide receptor B (NPR-B) regulates various physiological processes, including pituitary hormone secretion, cardiovascular stability, nervous system development, cell proliferation and bone growth. In this study, the full-length cDNA of Gallus gallus domesticus NPR-B gene was reported and the tissue expression of NPR-B gene in adult chickens was detected by quantitative real-time PCR (qPCR). The results showed that the full length of chicken NPR-B gene was 3 189 bp encoding a protein of 1 062 amino acids. The amino acid sequence of NPR-B was highly homologous to that of Homo sapiens (79%), Rattus norvegicus (78%), Xenopus laevis (73%) and Danio rerio (67%). The result of qPCR showed that NPR-B gene was highly expressed in the heart, muscle, midbrain and pituitary of chicken.
