Hippo信号通道中Lats2基因表达与湖羊肌肉生长发育的关系

Abstract:

[目的]探究Hippo信号通道中Lats2基因与湖羊肌肉生长发育的关联性。[方法]以湖羊作为试验材料,用RT-qPCR技术分析Lats2基因在不同性别、不同生长阶段(2、60、180日龄)、不同肌肉组织(比目鱼肌、背最长肌、腓肠肌和趾长伸肌)的时空表达规律,及其与Lats1、YAP1(Yes相关蛋白1)基因和肌肉生长相关基因在不同生长阶段的相关性。[结果]根据Hippo信号通道中Lats2基因的时空表达规律可以发现:在不同肌肉组织中,Lats2基因在趾长伸肌中相对表达量最高;在不同生长阶段,Lats2基因在60日龄的表达量最高;在不同性别中,Lats2基因的表达表现为公羊高于母羊。相关性分析结果表明:在2日龄肌肉组织中,Lats2基因的表达与MSTN基因间正相关(P<0.05);在60日龄的肌肉组织中,Lats2基因的表达与Lats1基因正相关(P<0.05);在180日龄肌肉组织中,Lats2基因的表达与MyoG基因间极显著正相关(P<0.01),与YAP1基因间显著负相关(P<0.05)。[结论]Hippo信号通道中Lats2基因的表达受不同性别、年龄和肌肉组织的影响,可能通过下调YAP1基因的表达抑制肌肉生长发育,可作为肌肉生长发育调控研究的候选基因。
[Objectives] The experiment mainly explored the correlation between large tumor suppressor kinase 2 gene(Lats2)in Hippo signal channel and Hu sheep muscle growth by RT-qPCR technique. [Methods] In this study, we analyzed the spatial and temporal expression regulation of Lats2 in Hu sheep with the affection of different gender, growth stages(2, 60 and 180 day-old)and skeletal muscle tissue(soleus muscle, longissimus doris muscle, gastrocnemius muscle, extensor digitorum longus). The correlation of expression in different growth stages within large tumor suppressor kinase 1 gene(Lats1), Yes-associated protein 1 gene(YAP1)and genes, which could affect the growth of muscle tissue(MyoG, MyHCⅠ, MSTN, MyHCⅡA, MyHCⅡX), or the genes related to muscle growth was emphatically discussed. [Results] It suggested that Lats2 in Hippo signal channel had the highest related expression in the extensor digitorumlongus muscle on the analysis of different muscle tissue expression. Affected by different growth stages, Lats2 gene had the highest related expression in the 6-month-old, and higher in the 2-day-old and the 2-month-old. In different genders, the related expression of rams was higher than that of ewes. On the analysis of related expression of Lats2, Lats1, YAP1 and genes that could affect the growth of muscle tissue, we found that Lats2 in 2-day-old muscle tissue had significant positive correlation with MSTN(P<0.05). In 2-day-old muscle tissue, there was a significant positive correlation between Lats2 and Lats1(P<0.05). The expression of Lats2 and MyoG had highly significant positive correlation(P<0.01), and the correlation of the expression between Lats2 and YAP1 was significant negative in 6-month-old muscle tissue(P<0.05). [Conclusions] The expression of Lats2 gene in Hippo signal channel was affected by different gender, growth stages and skeletal muscle tissue. We can down-regulate the expression of YAPA1 to inhibit the muscle growth, which can be a candidate gene of the growth of muscle fiber
