金华火腿粗肽液对PC12细胞氧化损伤的保护作用

Abstract:

[目的] 本文旨在研究金华火腿粗肽液对过氧化氢诱导大鼠嗜铬细胞瘤细胞(PC12)氧化损伤的保护作用。[方法] 以金华火腿粗肽液为研究对象,建立PC12细胞过氧化氢损伤模型,测定不同质量浓度(200和400 μg?mL-1)粗肽液对氧化损伤细胞的存活率和细胞中丙二醛(MDA)含量、乳酸脱氢酶(LDH)渗漏率及过氧化氢酶(CAT)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性的影响。[结果] 经过尺寸排阻色谱分离的金华火腿粗肽液CPC,显著提高了受损细胞的存活率,同时提升了细胞中的CAT、SOD和GSH-Px酶活性,降低了细胞中MDA含量和LDH渗漏率,其中以400 μg?mL-1粗肽液处理组效果最显著。[结论] 金华火腿粗肽液对过氧化氢引起的PC12细胞损伤具有保护作用。
[Objectives] The paper aims to study the protective effects of Jinhua ham peptide extract on alleviating oxidative damage induced by hydrogen peroxide(H2O2) in PC12 cells.[Methods] A cell model of oxidative damage was established by H2O2. PC12 cells were treated with different concentrations of crude peptides. The cell viability,malondialdehyde(MDA) content,the leakage rate of lactic dehydrogenase(LDH) and catalase(CAT),superoxide dismutase(SOD),and glutathione peroxidase(GSH-Px) activity in these cells were measured.[Results] Crude peptide C(CPC) presented strong scavenging activity against superoxide anion radicals in vitro. Incubating cells with different concentrations of CPC enhanced the cell viability and antioxidant enzyme activities including CAT,SOD and GSH-Px,while decreased the leakage rate of LDH and cellular lipid oxidation. These cytoprotective effects of CPC on PC12 cells showed a significant dose response relationship. And the treatment of CPC(400 μg?mL-1) had the most significant effect.[Conclusions] Crude peptides extracted from Jinhua ham has protective effects on PC12 cell injury induced by H2O2
