榉树枯萎病菌毒素的提取及生物活性的测定DOI: 10.13360/j.issn.1000-8101.2015.01.029 Keywords: 榉树枯萎病, 尖孢镰刀菌, 毒素, 生物活性the blight disease of Zelkova, Fusarium oxysporum, toxin, biological Abstract: 为了明确榉树枯萎病菌毒素的致病能力,对榉树枯萎病菌毒素进行了提取及生物活性的测定。结果表明:病原菌毒素滤液的生物活性主要体现在对于榉树胚根生长的抑制作用以及对于榉树幼苗的致萎性。通过对培养液种类、培养温度、培养时间、培养方式、培养环境酸碱度以及光照与否6项指标进行筛选,最终确定使用改良Czapek培养液,在25 ℃下,持续光照,连续振荡,pH为7的条件下可以获得较多的毒素。毒素对榉树的胚根生长存在明显的抑制作用,毒素的浓度越高,抑制作用就越明显。不同浓度的毒素滤液处理对榉树幼苗的致萎作用不同,浓度越高致萎性也越强。The blight disease of Zelkova is a new disease which occurred in Maoshan area of Jurong City in Jiangsu Province in recent years. The outbreak of this disease has led to a huge loss on propagation of Zelkova for several years. Extraction and biological test of toxin in the blight disease of Zelkova had been carried out. The toxin of the pathogen depressed the plant's radicle growth and led to seedling shrivel up. By the screening on culture liquids, temperature, time, model, pH of liquids and sunlite or not, it was found that the optimal culture medium for toxin production was new Czapek liquid, and the optimum culture condition was 25 ℃,light, continuous shock, pH 7. Higher concentration of toxic substance presented higher possibility to depress the plant's radicle growth and cause blight of seedling
