Abstract:

] 目的 · 探讨重组脂联素对自然流产(spontaneous abortion,SA)模型小鼠妊娠结局及 Th17/ 调节性 T 细胞(Treg)平衡的影 响。方法 · 将 CBA/J 雌鼠与 DBA/2 雄鼠交配建立 SA 模型,CBA/J 雌鼠与 BALB/c 雄鼠交配建立正常妊娠模型,于妊娠第 5 日起脂联 素组连续腹腔注射重组脂联素 10 μg/(kg · d)×9 d,对照组注射等量 PBS。于妊娠第 14 日处死孕鼠,计算胚胎吸收率;ELISA 检测血 清脂联素、白细胞介素 17(IL-17)和转化生长因子 β(TGF-β);分离脾脏淋巴细胞,流式细胞术检测 Th17/Treg 细胞分群。结果 · 重 组脂联素对正常妊娠模型小鼠胚胎吸收率无明显影响,但可使 SA 模型小鼠胚胎吸收率由 32.2% 降至 13.3%(P=0.039);ELISA 结果 显示,SA 模型小鼠腹腔注射脂联素后血清脂联素水平显著升高,IL-17 水平降低,TGF-β 水平升高(均 P<0.01);流式细胞术结果显 示,脂联素可降低 SA 模型小鼠 Th17/CD4+ T 细胞比例,升高 Treg/CD4+ T 细胞比例,逆转 Th17/Treg 比值。结论 · 在孕早期腹腔注射 重组脂联素可改善 SA 模型孕鼠的妊娠结局,可能与其对 Th17/Treg 平衡的调控有关。
:Objective · To investigate the effect of recombinant adiponectin on the pregnancy outcome and the balance of Th17/regulatory T cell (Treg) in spontaneous abortion murine model. Methods · CBA/J(♀)×DBA/2(♂) mating was used as a spontaneous abortion model, while CBA/J(♀)×BALB/ c(♂) mating as a normal pregnancy model. CBA/J female mice were intraperitoneal injected with 10 μg/(kg·d) recombinant adiponectin for 9 d from day 5 of gestation. The control group received equal volumes of PBS each day. On day 14 of gestation, the mice were killed. The embryo resorption rate was calculated. The levels of serum adiponectin, IL-17 and TGF-β were detected by ELISA. Spleen lymphocytes were isolated. The ratios of Th17/Treg were counted by flow cytometry(FCM). Results · Recombinant adiponectin administration had no obvious effect on the embryo resorption rate of the normal pregnancy model, while it significantly reduced the embryo resorption rate from 32.2% to 13.3% (P=0.039) of the spontaneous abortion model. ELISA showed that recombinant adiponectin increased the serum adiponectin and TGF-β levels, and reduced the IL-17 level of the spontaneous abortion model (P<0.01). FCM showed that adiponectin administraion could reduce the ratio of Th17/CD4+ T cells, increase the ratio of Treg/CD4+ T cells and reverse the ratio of Th17/Treg in abortion mice. Conclusion · Intraperitoneal administration of recombinant adiponectin in early gestation can improve pregnancy outcome in a murine model of abortion maybe through regulating the balance of Th17/Treg
