‘里昂城’铁线莲组培体系的建立Establishment of Regeneration System of Clematis ‘Ville de Lyon’Abstract: 以2年生铁线莲园艺品种‘里昂城’带芽茎段为外植体材料,建立了完整的组织培养体系。结果表明,‘里昂城’铁线莲带芽茎段较适宜的灭菌方法是0.1% HgCl2二次灭菌6 min和5 min,污染率控制为5.56%;初代培养诱导腋芽的最佳培养基为MS+1.0 mg/L 6-BA+0.1 mg/L NAA,出芽率为93.33%,出芽指数为1.70;MS+1.0 mg/L 6-BA+0.1 mg/L NAA+1.0 mg/L KT 是芽增殖的最佳培养基,增殖系数高达4.07,不定芽生长良好;最佳生根培养基为1/2MS+0.3 mg/L NAA+3.0 mg/L IBA,生根率高达93.33%;生根苗移栽至珍珠岩和草炭土(1∶1,V/V)混合基质中成活率为73.00%。In vitro regeneration protocol from excised stems of Clematis ‘Ville de Lyon’ via direct method was developed.The results differed significantly among the various experimental treatments,and low contamination (5.56%) and high germination rate (78.89%) were achieved when stem segments were disinfected with 0.10% HgC12 twice (6 and 5 min);The suitable initial culture medium was Murashige and Skoog (MS) medium supplemented with 1.0 mg/L 6-Benzylaminopurine (6-BA) and 0.1 mg/L 1-Naphthaleneacetic acid (NAA),in which the germination rate was 93.33% with a shoot forming index of 1.70.Proliferation rate adventitious bud on MS medium complemented with 1.0 mg/L 6-BA,0.1 mg/L NAA and 1.0 mg/L Kinetin (KT) was significantly higher (P<0.05) than the other treatments,in which proliferation coefficient was 4.07.Adventitious buds were rooted well on half strength MS medium (1/2MS) with 3.0 mg/L Indole-3-butytric acid (IBA) and 0.3 mg/L NAA,and the rooting ratio was 93.33%.The survival rate of seedlings was 73.00% in the substrate mixing of perlite and turfy soil as the rate of 1∶1 (V/V).
