运用氨基酸指纹图谱研究牛乳产地和季节变化Differentiating Geographical and Seasonal Variations of Milk Production Using Ultraperformance Liquid Chromatography Amino Acid Fingerprints Combined with Chemometrics AnalysesAbstract: 本文通过超高效液相色谱(ultraperformance liquid chromatography,UPLC)氨基酸指纹图谱融合化学计量学的方法对来自35个牧场的361个牛乳样品进行地理位置区分和季节区分。以15种氨基酸含量与谷氨酸含量的比率为变量,利用Matlab软件进行主成分分析及偏最小二乘判别分析。结果表明,Lys/Glu和Ile/Glu可作为牛乳样品地理位置区分及山东地区样品季节区分的特征变量。偏最小二乘判别分析建立的模型用于山东省与上海市及苏州市的地理位置区分、上海市及苏州市样品的季节区分及山东省样品的季节区分的预报值正确率分别为83.3%,72.7%和91.7%。UPLC氨基酸指纹图谱融合化学计量学方法为牛乳溯源提供了重要的依据。The geographical and seasonal variations of 361 milk samples produced from 35 farms were differentiated by ultraperformance liquid chromatography (UPLC)amino acid fingerprints combined with chemometrics analyses.Fifteen averaged amino acid mole ratios (the concentration of each amino acid to glutamic acid)were used as variables.Principal component analysis (PCA)and partial least-squares-discriminant analysis (PLS-DA)were used by MATLAB.Results showed that the mole ratios of Lys and Ile to Glu were characteristic variables for geographical variation and seasonal variation at Shandong province.The correct classification rate for milk samples of different geographical conditions was 83.3%.The correct classification rates for seasonal variations at Shanghai,Suzhou cities and Shandong province were 72.7% and 91.7%,respectively.The results indicated that UPLC amino acid fingerprints combined with chemometrics analyses may be useful on the research of origin of milk and its related products.
