黄瓜不同性型花中差异表达的MADS-box基因MADS-Box Genes Differentially Expressed in Diverse Sex Types in Cucumber(Cucumis sativus L.)Abstract: MADS-box基因家族是一类转录调控因子,影响植物各个生长发育的环节,尤其是花器官的发育。随着模式植物MADS-box基因研究的深入,一些该类基因的作用方式已经被阐明。黄瓜的性型直接关系到其产量,黄瓜中MADS-box基因对其性型的影响尚未清楚。本研究对6组不同黄瓜性型的近等基因系材料的花芽以及顶芽进行转录组分析,利用生物信息学方法,获得9个不同性型花芽和顶芽差异表达的MADS-box基因:Cucsa.018420、Cucsa.113420、Cucsa.251170和Cucsa.327970在雄花中特异表达,Cucsa.139620、Cucsa.160640、Cucsa.241990在雌花和两性花中的表达差异显著,Cucsa.212720和Cucsa.392160在雌花中特异表达。通过荧光定量RT-PCR验证了其中两个差异表达基因Cucsa.018420和Cucsa.392160的表达,发现两者在整个花发育过程中的表达规律与转录组结果一致。本研究初步探讨了MADS-box基因对黄瓜花性型的影响,为黄瓜MADS-box基因的深入研究提供参考。The MADS-box gene family is a kind of transcription factors which affect growth and development of plants,especially the floral organ development.With the research of MADS-box genes in model plants,their functions have been elucidated.The functions of MADS-box genes on sex-type of cucumber are unravelled,which is important for cucumber production.Six pairs of near isogenic lines of cucumber with different sex types were used for transcriptome analysis in this study.There are 9 MADS-box genes differentially expressing in floral buds and apical buds:Cucsa.018420,Cucsa.113420,Cucsa.251170 and Cucsa.327970 expressed specifically in male flowers;Cucsa.139620,Cucsa.160640 and Cucsa.241990 differentially expressed in female flowers and perfect flowers; Cucsa.212720 and Cucsa.392160 expressed specifically in female flowers.With real-time quantitative reverse transcription PCR,the expression patterns of Cucsa.018420 and Cucsa.392160 in floral budswere verified,which were consistent with the transcriptome analysis results.This research on MADS-box genes affecting sex type of cucumber should provide a reference for uncovering their function in cucumber.
