Abstract:

为了揭示蛋白质的差异表达和C4植物杂交狗牙根叶片和根系对高温抗性差异的关系,研究了0、1、6、24 d的高温胁迫对杂交狗牙根(‘Tifdwarf’)叶片和根系抗热相关生理指标和可溶性蛋白差异表达的影响。通过草坪质量、光化学效率、叶绿素和类胡萝卜素含量的分析,发现与1、6 d的高温胁迫相比,24 d高温胁迫条件下,叶片的伤害显著增加。通过电解质外渗分析发现,与叶片相比,高温对根系的伤害更大。结合标准蛋白,笔者发现叶片中热激蛋白(heat shock protein,Hsp),Hsp90、Hsp83、Hsp75、Hsp68、Hsp60、Hsp49,根系中Hsp90、Hsp75、Hsp68、Hsp60的差异表达,可能与杂交狗牙根叶片和根系对高温抗性的差异有关。研究结果对于筛选C4植物抗高温的分子标记以及植物根系蛋白质组学的研究具有重要意义。
To clarify the relationship between the differential protein expression and heat tolerance of leaves and roots in the C4 plant hybrid bermudagrass(Cynodon transvaalensis×Cynodon dactylon,‘Tifdwarf’),the influences of 0,1,6 and 24 d of heat stress on related physiological indexes and the differential expression of soluble proteins was analyzed.Compared with 1 d and 6 d of heat stress,heat damage increased significantly at 24 d of heat stress through analysis of turf quality,photochemical efficiency,chlorophyll and caroteonoid content.It was found that the differential expressions of heat shock proteins(Hsp),including Hsp90,Hsp83,Hsp75,Hsp68,Hsp60 and Hsp49 in leaves,and Hsp90,Hsp75,Hsp68 and Hsp60 in roots,were associated with different heat tolerance between leaves and roots,compared with the protein marker.Results from this study might be useful for screening molecular markers for heat tolerance in C4 plants as well as proteomic studies of roots in all plants.
