下丘脑室旁核中CART 55-102在创伤应激诱导的大鼠饮酒行为中的作用Keywords: traumatic stress drinking behavior CART 55-102 rat Abstract: Objective To observe the changes of drinking behavior after traumatic stress in adolescent rats,and explore the role of cocaine-and-amphetamine-regulated transcript (CART) 55-102 of the hypothalamic paraventricular nucleus (PVN). Methods Viscera exploration surgery was simulated to establish traumatic model in adolescence rats.Male SD rats were randomly divided into two groups (n=6):control and trauma group,and measured the drinking behavior (including alcohol intake and preference) during 14 days by 2-bottle choice drinking paradigm and the level of ethanol in serum.The immune activity of CART 55-102 in the PVN was detected through immunohistochemical staining.The activity of CART 55-102 in the PVN was intervened by a stereotactic location method in traumatic rats injected by saline and CART 55-102 (n=6).The drinking behavior was measured to explore the role of CART 55-102 in drinking behavior induced by traumatic stress. Results Compared with control rats,alcohol intake,(g*kg-1*d-1) alcohol preference (%) and blood ethanol concentration (mg/dL) in traumatic rats increased statistically (2.31±0.08 vs. 1.76±0.19,16.79±0.53 vs.12.47±1.61,16.78±2.19 vs. 8.85±1.69,all P<0.05),and the immune activity of CART 55-102 in the PVN in traumatic rats decreased statistically (0.76±0.07 vs. 1.00±0.09,P<0.05).Further,CART 55-102 injection into the PVN decreased alcohol intake (1.05±0.12 vs. 1.98±0.13,P<0.001),alcohol preference (6.78±0.61 vs. 10.13±0.73,P<0.01),and blood ethanol concentration (16.64±3.28 vs. 29.83±3.82,P<0.05) compared with saline group. Conclusions Traumatic stress can stimulate drinking behavior in adolescent rats,possibly through inhibiting the immune activity of CART 55-102 in the PVN.
