Abstract:

摘要 目的: 观察ERK5抑制剂对巨噬细胞胞葬作用及相关信号分子ProS、Axl表达的影响.方法: 体外常规培养RAW264.7巨噬细胞,并用ERK5抑制剂(XMD8-92和ERK5-IN-1)干预细胞,实验分为正常对照组、XMD8-92组和ERK5-IN-1组;流式细胞仪检测巨噬细胞的胞葬率,RT-qPCR和Western blot法检测巨噬细胞ProS和Axl的mRNA和蛋白的表达.结果: 与正常对照组相比,XMD8-92组和ERK5-IN-1组巨噬细胞胞葬率下降(P<0.05),ProS和Axl的mRNA和蛋白的表达水平下调(P<0.05).结论: ERK5抑制剂可以抑制巨噬细胞的胞葬作用,下调胞葬作用相关信号分子ProS和Axl的mRNA和蛋白的表达,提示ERK5激酶的失活可能通过下调ProS、Axl的表达导致巨噬细胞胞葬作用受损.
