Abstract:

摘要 目的: 用生物信息学方法探讨与非创伤性股骨头坏死相关的潜在关键基因和信号通路. 方法:GEO数据库中下载得到8例人股骨头样本产生的GSE74089表达谱数据进行生物信息学分析,Limma包筛选差异表达基因.用DAVID数据库、R包、STRING数据库及Cytoscape软件对差异表达基因进行基因功能、信号通路及蛋白-蛋白相互作用网络分析.结果:共筛选出1 315个差异表达基因,上调基因809个,下调基因506个.差异表达基因主要涉及细胞外基质、胞外空间、胶原蛋白三聚体、胶原纤维组织、松弛素信号通路及转化生长因子β信号通路等. PPI网络由391个节点和1 213个交互组成,网络分析列出蛋白互作网络的前20个中心节点蛋白,挖掘出4个潜在关键基因.结论: HACE1、FBXW7、GNG8和SAA1等差异基因可能与非创伤性股骨头坏死的发病机制密切相关.
