云南保山温泉产纤维素酶高温菌多样性研究

Abstract:
为了探寻温泉中的未知菌种,寻找能用于工业生产的产酶菌株,分别对云南省保山市腾冲县和昌宁县温泉中产纤维素酶高温菌进行筛选分离鉴定,并研究其多样性.采用不同的温度和ph培养条件,从温泉中筛选出具有纤维素分解能力的菌株200株.通过对菌株的16srrna基因序列测序鉴定分析,确定其分属于8个属30个种.在55℃培养条件下分离获得菌株122株,分别属于8个属26个种.在65℃条件下分离获得菌株78株,分别属于5个属15个种.菌株纯化转接4代后检测产酶活性,95%以上产纤维素酶的菌株同时具有淀粉酶的分解能力,50%左右的产纤维素酶菌株同时产蛋白酶.研究表明在腾冲温泉和昌宁温泉中,存在着较多产纤维素酶、淀粉酶、蛋白酶的菌株.图3表3参21
