海洋生防细菌在黄瓜根际土壤定殖规律的研究*

Abstract:
【目的】本文的目的是揭示土壤初始ph值、温度和有机质含量三因素对多粘类芽孢杆菌(paenibacilluspolymyxa)l1-9菌株在黄瓜根际土壤定殖的影响。【方法】本文采用室内盆栽实验,在黄瓜一叶期灌根接种生防菌l1-9发酵液,设置土壤ph值、温度和有机质含量的不同处理,培养并定期采集黄瓜根际土壤样品,采用实时荧光定量pcr技术检测了海洋生防菌l1-9在黄瓜根际土壤定殖的数量变化。【结果】通过定量数据及对其显著性检验结果表明,实时荧光定量pcr技术适用于研究土壤根际生防菌的定殖。温度、初始ph值和有机质含量在一定时间内显著影响l1-9菌株在黄瓜根际土壤中的定殖。土壤初始ph为6、7和8均可定殖。初始ph值为7和8的处理,在多数调查时间内无显著差异。在存在显著差异的时间内,初始ph值为6时,l1-9菌株在黄瓜根际土壤中定殖数量在同期中最大。15℃培养时,l1-9菌株能够在黄瓜根际土壤中较长时间内保持较高定殖数量。温度越高,定殖越早,但在较高温度下定殖数量下降较快。高有机质含量可以增加定殖数量,提高定殖能力,但是有可能减缓定殖速度。【结论】海洋生防菌l1-9能够在较宽温度、初始ph值和有机质含量范围内稳定定殖,较低温度、中性偏酸初始ph值和高有机质含量有利于l1-9菌株在黄瓜根际土壤中定殖。图1表3参18
