东亚飞蝗mariner转座子片段的克隆和序列分析

Abstract:
mariner转座子在昆虫中广泛存在,转座时不需要宿主特异因子的作用,是昆虫种系转化的有力工具.以mariner转座酶的保守序列设计的简并引物从东亚飞蝗locustamigratoriamanilensis(meyen)基因组中扩增出7个序列有差异的片段,其中的1个片段能够翻译成为连续的氨基酸序列,另外6个片段中均含有终止子或移码突变.经同源性比对、序列特征分析,证实东亚飞蝗基因组中存在mariner类元件,但大部分都是无功能的假基因;系统进化分析发现东亚飞蝗基因组中的mariner类元件有可能属于新的mariner次级家族.图3参33
