麻疯树叶片高效再生体系与不定芽起源

Abstract:

建立了培养周期短、再生效率高的麻疯树叶片再生体系.6-ba是诱导麻疯树叶片产生愈伤组织的关键外源激素,添加低浓度的2,4-d能增强6-ba的诱导效果.最佳愈伤组织诱导培养基是ms+2.0mg/l6-ba+0.05mg/l2,4-d,瘤状愈伤组织的诱导率高达91.98%±2.1%.光培养下产生的愈伤组织质地紧密,瘤状结构丰富,再生能力强.随着继代次数的增加,愈伤组织褐化加重,逐渐失去再生能力.不定芽再生培养基中,比较了3种细胞分裂素6-ba、kt和tdz对诱导愈伤组织分化不定芽的影响.6-ba诱导不定芽的能力最优,其次是tdz,kt最差.kt与ba的组合诱导不定芽再生的能力最强,最适的不定芽再生培养基为ms+0.5mg/l6-ba+0.5mg/lkt+0.1mg/liba+0.1mg/lga3,再生率高达68.01%±2.5%,平均每个愈伤组织分化出4.25个芽.70%的再生苗在ms培养基添加0.2mg/liba或naa(0.1或0.2mg/l)中均能产生根系,平均根系5.2个.对麻疯树叶片再生的各阶段进行了石蜡切片,揭示了不定芽的起源及发育的全过程,明确了不定芽起源于叶片的表皮细胞层.图3表3参30
