黄曲霉培养条件的优化及黄曲霉毒素b1的提取

Abstract:

为提取高浓度的黄曲霉毒素b1,以大米为培养基培养黄曲霉,分别在不同温度梯度(15℃、20℃、25℃、30℃、35℃)和湿度梯度(0%、80%、85%、90%和95%和100%)下培养,每天测量一次菌斑直径的变化(共测18次),然后采用logistic(一级)和ratkowsky(二级)模型对黄曲霉菌斑大小进行拟合和检验,最后借助响应面模型对黄曲霉产毒条件进行优化.结果显示,在35℃和100%湿度的培养条件下,只需培养16~17d即可获得直径较大的黄曲霉菌斑,提取黄曲霉毒素b1浓度为3312ng/ml,单位面积黄曲霉毒素b1产量是优化前(1101.471ng/ml)的3倍.图5表4参18
