白囊耙齿菌腐朽木材相关cdna片段的序列克隆与分析

Abstract:

为了解白囊耙齿菌腐朽木材的分子机制,利用基因差异显示技术(differentialdisplayreversetranscriptase,pcrddrt-pcr)研究了白囊耙齿菌腐朽白桦木材前后基因表达的差异.共分离出5个腐朽后在菌丝体中特异表达的cdna片段,分别命名为a2、a7、c6、b8和b3.序列同源性比对发现:片段a2、a7和c6分别与编码漆酶(laccase)、纤维素酶a(cellulasea)和麦芽糊精磷酸化酶(maltodextrinphosphorylase)等3个基因具有高度同源性;b8与一个编码响应调节蛋白的基因具有高度同源性.研究结果显示,所筛选的差异表达基因a2、a7和c6等与木材降解有密切关系.图2表2参22
