不同窖龄窖泥古菌的系统发育多样性分析

Abstract:

为探索不同窖龄窖泥古菌类群与演替规律,分别提取20、50和150年窖龄窖泥样品总dna,应用pcr-ardra和16srdna克隆测序技术对不同窖龄窖泥古菌群落结构进行研究.结果表明:191个克隆子分成22个可操作分类单元(otus),系统发育分析归为6个类群,分别属于甲烷八叠球菌属(methanosarcina)、甲烷杆菌属(methanobacterium)、甲烷袋状菌属(methanoculleus)、甲烷粒菌属(methanocorpusculum)、甲烷鬃菌属(methanosaeta)和甲烷螺菌属(methanospirillum).不同窖龄窖泥的优势古菌类群有明显差异,20年窖泥优势古菌类群以乙酸利用型methanosarcina、methanosaeta为主,50年、150年龄窖泥优势古菌类群以氢气营养型methanocorpusculum、methanobacterium为主;随着窖龄的增加,窖泥古菌shannon-wiener多样性指数(h)和berger-parker优势度指数(d)呈增加的趋势,pielou均匀度指数(j)呈降低趋势;不同窖龄窖池古菌的相似性系数为0.429~0.677,50年和150年窖泥的菌群结构最为相似.本研究揭示了不同窖龄窖泥古菌系统发育多样性的差异与窖泥老熟的关系.图1表1参22
