茎瘤芥pr4基因的克隆与表达分析

Abstract:

病程相关蛋白(prs)在植物抗病过程中起着重要作用,pr4是prs家族中参与植物系统获得抗性的重要效应因子.利用race技术从受十字花科根肿菌plasmodiophorabrassicae侵染的茎瘤芥中克隆到一个pr4基因,命名为bjpr4(genbank登录号为jx467703),bjpr4基因核苷酸序列全长584bp,编码140个氨基酸;bjpr4蛋白具有高度保守的barwin结构,与其它植物的pr4蛋白具有较高的相似性,系统进化分析表明该蛋白基因在不同物种之间具有保守性.bjpr4序列中没有几丁质结合域结构,所以这个基因属于ⅱ类pr4.qrt-pcr检测分析显示,十字花科根肿菌处理后,bjpr4在病程初期表达量较低,病程后期表达量急剧上升;水杨酸处理后,bjpr4在短期内上调表达,12h表达量最高,随后降低至较低水平;而茉莉酸甲酯、乙烯、脱落酸处理后该基因表达水平均下降.研究结果表明bjpr4在茎瘤芥根部的表达受到不同植物信号分子的调控,bjpr4在根肿病病程中的表达特征表明bjpr4与茎瘤芥对根肿菌的抗性有关.
