孔雀石绿和结晶紫在荧光检测中的比较

Abstract:

为优选出三苯甲烷染料中的一种染料与g-四聚体结合开发为荧光定量报告体系,对孔雀石绿(malachitegreen,mg)和结晶紫(crystalviolet,cv)与g-四聚体结合后的灵敏度和重现性进行了比较研究.首先,比较了4种不同dna(hum21,short-hum21,double-strands,single-strand)分别与孔雀石绿和结晶紫结合后的荧光光谱,结果显示,两种染料均可区分以上4种dna,但孔雀石绿的灵敏度更高.然后,将孔雀石绿和结晶紫应用到实时荧光检测中,进一步比较验证两种染料灵敏度,并考察两种染料的重现性,结果显示,孔雀石绿的重现性和灵敏度均优于结晶紫.由此,建立了孔雀石绿与g-四聚体结合的荧光定量报告体系.图4表1参21
