骨髓间质干细胞移植后大鼠缺血心肌的超微结构观察

Abstract:

目的观察骨髓间质干细胞(mscs)移植后大鼠缺血心肌组织的超微结构改变。方法体外培养、纯化sd大鼠的骨髓mscs,4’6-二脒-2-苯基吲哚(dapi)标记,经尾静脉注射移植治疗大鼠急性心肌缺血,移植后1周和8周荧光显微镜下检测心肌组织中dapi阳性细胞,透射电镜观察心肌组织的超微结构改变。结果mscs经传代培养至第3代后,均一性好。细胞移植后1周和8周,移植组的心肌组织中可见呈蓝色细胞核的dapi阳性细胞,而对照组未发现阳性细胞。移植后1周,心肌组织的超微结构观察发现移植组的心肌梗死周边区域可见少量细胞,其形态类似于体外培养的骨髓mscs;移植后8周,移植组梗死周边可见大量的毛细血管,其内皮细胞细胞“芽生”分割管腔开始形成新的血管,还可见少量“不成熟”的心肌细胞。结论骨髓mscs通过血液循环可向缺血心肌组织归巢并促进血管和心肌组织的再生。
