新基因lx3与白细胞介素-6诱导的相关性

Abstract:
目的研究功能未知新基因lx3[1]与白细胞介素-6(il-6)的相关性,为研究il-6的作用机制寻找新的靶基因。方法反转录-pcr(rt-pcr)分析不同浓度il-6处理的u937细胞及同一浓度il-6诱导不同时间的u937细胞中新基因lx3的表达差异;northernblot分析il-6诱导前后u937细胞中新基因lx3表达的变化。结果lx3基因在u937细胞中的表达量随il-6诱导浓度的增加而增加,在il-6浓度为500ng/ml时表达量最高,而在0ng/ml时不表达;时间表达谱分析显示在il-6刺激8h时新基因lx3的表达量最高;northern印迹分析显示新基因lx3在il-6诱导后的u937细胞中的表达量明显升高。结论lx3是与il-6诱导紧密相关的新基因。
