氯沙坦对糖尿病大鼠肾组织mt3-mmp及timp2mrna表达的影响

Abstract:

目的观察糖尿病大鼠肾组织膜3型基质金属蛋白酶(mt3-mmp)及组织2型金属蛋白酶抑制物(timp2)mrna表达的变化及氯沙坦对它们的影响。方法建立糖尿病大鼠模型并将之分为正常对照组(a组)、糖尿病组(b组)、糖尿病+氯沙坦组(c组)。饲养18周后取出肾脏以rt-pcr法检测mt3-mmp、timp2及tgf-β1mrna的表达,电镜观察大鼠肾小球基底膜厚度及系膜基质密度(系膜基质面积/系膜面积),收集24h尿检测尿白蛋白。结果b组大鼠肾组织mt3-mmpmrna、timp2mrna和tgfβ1mrna表达以及均显著高于a组,c组显著低于b组,但与a组无显著差异。b组尿白蛋白、基底膜厚度及系膜基质密度显著高于a组,c组均显著低于b组。p<0.05。结论氯沙坦治疗可延缓糖尿病肾病的发生,并可抑制糖尿病大鼠肾组织mt3-mmp及timp2mrna表达。
