高低转移人大肠癌细胞系基因表达谱差异

Abstract:
目的用自制肿瘤转移相关基因cdna微阵列研究人大肠癌高转移细胞系lovo、sw620及低转移细胞系sw480、ls174t的基因表达谱差异,筛选与大肠癌转移相关的特异基因。方法分别提取4种大肠癌细胞系及对照的正常大肠上皮组织的mrna,逆转录成cdna探针,经cy3和cy5标记后分别与含有399个肿瘤转移相关基因的微阵列杂交,用专用软件分析杂交信号强度。结果高转移细胞系与低转移细胞系的基因表达谱有显著差异,其中有104个基因的表达值有意义,包括上调基因44个、下调基因60个。结论大肠癌转移是由其发生过程中多个基因表达的复杂变化决定的,高转移细胞系与低转移细胞系比较存在差异的基因可能与高转移特性有关。
