单胺氧化酶单克隆抗体对大鼠血管源性脑水肿的保护作用

Abstract:
目的观察单胺氧化酶单克隆抗体(maomab)对大鼠血管源性脑水肿(vbe)的治疗作用。方法75只wistar大鼠随机分为无脑水肿组、脑水肿未治疗组、脑水肿生理盐水组、脑水肿甘露醇组与脑水肿maomab组。用腹腔注射苯肾上腺素的方法制成vbe模型,然后分别注射生理盐水、甘露醇或maomab到大鼠腹腔,通过远红外线水分分析仪分别测定各组脑灰、白质水分含量百分比。用evansblue(eb)测定血脑屏障(bbb)的通透性。结果maomab对减少vbe大鼠脑灰、白质水分含量、降低bbb通透性均有显著效果(p<0.01),尤其对白质的脱水作用更显著(p<0.01),而与无脑水肿组想比无显著差异(p>0.05)。甘露醇组也可降低灰质、白质水肿,但对灰、白质无特异性差异,降低bbb通透性差。结论vbe中bbb的通透性改变与单胺氧化酶活性有关,maomab对vbe尤其对脑白质水肿有选择性治疗作用。
