plunc序列启动子转基因小鼠模型的建立

Abstract:
目的利用携带有鼻咽组织相对特异性启动子plunc构建的载体建立转基因动物模型。方法将plunc-egfp质粒用xhoⅰ酶切线性化,胶回收线性化片段,稀释浓度4μg/ml。采用显微注射法将外源基因注射入小鼠受精卵细胞内。结果产13只小鼠。pcr检测基因整合,其中阳性12只,阳性率92.30%;southernblot检测阳性3只,阳性率23%。结论鼻咽组织相对特异性启动子plunc可有效地将外源基因整合入小鼠体内。
