基因芯片技术分析系统性红斑狼疮患者外周血白细胞基因表达谱的初步研究

Abstract:
目的比较系统性红斑狼疮(sle)患者与正常对照外周血白细胞的基因表达谱的改变,筛选sle差异表达基因。方法采集静脉血5ml提取总rna后,反转录合成、标记cdna探针,与基因芯片杂交后检测。结果9例患者与正常人对照均有相似差异表达的基因共89个,涉及细胞因子及受体相关基因、免疫相关基因、细胞信号和传递蛋白、蛋白翻译合成、离子通道和运输蛋白、凋亡相关基因、dna和rna结合、转录蛋白、细胞的外基质成分等。聚类分析结果显示在不同病人间尽管存在个体差异性,但是在其基因表达变化模式上仍然具有极大的相似性。结论基因芯片技术筛选出的差异表达基因可能为进一步研究sle的发生和发展以及寻找分子诊断标志和药物治疗靶标提供线索。
