vegf启动子介导双自杀基因重组腺病毒对lovo细胞的体外杀伤作用

Abstract:
目的探讨vegf启动子驱动的cd/tk双自杀基因重组腺病毒(ad-vegfp-cdglytk)对大肠癌lovo细胞的体外杀伤作用.方法分别用重组腺病毒ad-vegfp-cdglytk、ad-cmv-cd、ad-cmv-tk、ad-cmv-cdglytk转染大肠癌lovo细胞,给予前药5-fc、gcv,测定lovo细胞的集落形成、细胞存活率及该治疗系统的旁观者效应.结果与ad-cmv-cd、ad-cmv-tk相比,ad-vegfp-cdglytk系统对lovo细胞的集落形成、细胞生长均具有更强抑制作用,并可见较明显的旁观者效应.而与cmv启动子的双自杀基因相比,其作用无显著性差异.结论vegf启动子驱动的双自杀基因治疗系统对大肠癌lovo细胞有确切的杀伤作用,其作用强度与强启动子cmv驱动的双自杀基因治疗系统相似.
