侧脑室注射链脲霉素引起大鼠脑aβ1-40、aβ1-42和tau202、tau396、tau404表达增加

Abstract:

目的探讨用链脲霉素(streptozotocin,stz)阻断脑内胰岛素信号传导,引起脑能量代谢障碍与aβ表达及tau蛋白过度磷酸化的关系。方法24只sd大鼠随机分为实验组和生理盐水组,实验组双侧侧脑室注入stz3mg/kg,第3天重复此剂量;生理盐水组手术操作相同,但注入等量的生理盐水代替stz。21d后,采用免疫组化方法观测各组大鼠大脑皮层和海马区的aβ1-40、aβ1-42、tau396、tau404阳性细胞表达。结果与生理盐水组相比,实验组皮层和海马aβ1-40、aβ1-42、tau202、tau396、tau404阳性细胞明显增多。结论侧脑室注射stz可以引起脑内aβ表达增多和tau蛋白过度磷酸化。
