抗吗啡抗体的制备及其特性评价

Abstract:

目的评价吗啡及琥珀酰化吗啡半酯两种交联物作为抗原免疫小鼠而产生的免疫效果及其中和阿片类物质的能力。方法吗啡及吗啡半酯在碳二酰亚胺催化下分别与bluecarrier载体蛋白交联,用其免疫小鼠,elisa检测免疫小鼠血清中抗体滴度及抗体6个月内效价维持状况,通过竞争性抑制实验检测抗体对吗啡及海洛因的体外结合能力,与其他阿片及非阿片类化学物质的交叉反应。结果两种交联物免疫小鼠第9周血清中平均抗吗啡抗体滴度均超过1:8000,其中m-6-s-blue组效价强度较m-blue组高,且效价维持时间较久;两组抗体对较低浓度吗啡及海洛因的抗血清抑制率均达50%以上,抑制率与吗啡或海洛因的浓度呈剂量依赖性。结论两种载体蛋白交联物均能诱导小鼠产生高滴度的抗吗啡抗体,其体内及体外实验观察均能产生中和吗啡的作用,提示其对控制复吸将有较好的应用前景。
