曲霉脲酶的动力学研究ⅰ

Abstract:
以脲素为底物,对曲霉脲酶的动力学特性进行了研究.在不同ph(5.0,5.6,6.0,6.6,7.0,7.6,8.0,8.6,9.0)条件下测定脲酶的km值和vmax.结果表明,ph对脲酶的vmax影响明显,在ph5.5至ph8.0之间vmax呈钟形.在ph7.0的酶促反应速度最快,km值不受ph值影响;不同的ph值(5.0,5.6,6.0,6.6,7.0,7.6,8.0,8.6,9.0)下脲酶的km值和vmax显示,在ph7.6时对底物的亲和力最大,为最佳ph,该脲酶在ph6.6到8.6时的稳定性较好.
